淫羊藿苷等6种中药单体抑制Cloudman S91黑素瘤细胞株黑素合成的研究 被引量:25

摘要 观察淫羊藿苷、白藜芦醇苷、红景天苷等6种中药单体对体外培养的CloudmanS91黑素瘤细胞黑素合成和酪氨酸酶活性的影响。方法:选择系列浓度梯度的中药单体作用于体外培养的CloudmanS91黑素瘤细胞,测定细胞酪氨酸酶活性、黑素含量和细胞增殖率。结果:淫羊藿苷、白黎芦醇苷和红景天苷3种药物明显抑制酪氨酸酶活性和黑素的生成(P<0.05),且呈浓度依赖性,作用3d内呈时间依赖性。其中淫羊藿苷的作用与氢醌比较,差异无显著性(P>0.05),而白黎芦醇苷和红景天苷的作用明显弱于氢醌(P<0.01)。淫羊藿苷、白黎芦醇苷和红景天苷均促进细胞的增殖,其中淫羊藿苷对细胞增殖促进作用明显。结论:淫羊藿苷、白黎芦醇苷和红景天苷能抑制CloudmanS91黑素瘤细胞黑素合成和酪氨酸酶活性。 Objective: To evaluate the effects of 6 kinds of monomers extracted from Chinese medicines including icariine?piceid and salidroside on tyrosinase activity and melanogenesis in Cloudman S91 melanoma cells. Methods: Different concentrations of 6 kinds of monomers were incubated with Cloudman S91 melanoma cells;then tyrosinase activity;melanin content and the rate of proliferation of melanoma cells were measured respectively. Results: Icariine; piceid and salidroside inhibited tyrosinase activity and melanin content in dose-dependent and time-dependent manner(P < 0.05). The effect of icariine of 50 μmol/L was the same as hydroquinone and effects of piceid and salidroside were weaker than those of the positive control.Three kinds of monomers could stimulate the proliferation of cells. Conclusions: Icariine; piceid and salidroside can inhibit tyrosinase activity and melanogenesis in Cloudman S91 melanoma cells.
