摘要
为了研究视黄醇结合蛋白 (RBP)在维生素A代谢调控中的作用和与相关疾病发生、发展的关系 ,以RBP作为诱饵基因构建了融合表达载体pGBKT7 RBP ;在排除自身转录激活活性的基础上 ,与人肾脏cDNA预转库融合筛选 (PretransformedMATCHMAKERLibraries) ,经营养缺陷型和X α gal显色鉴定 ,从中筛选出三种与RBP有相互作用的新蛋白质 。
On the basis of having constructed inducing fusion vector: pGBKT7-RBP, pretransformed human kidney cDNA library PACT2 and pGBKT7-RBP were co-expressed. After libraries screening and nutritional selection as well as X-α-gal analysis, three protein sequence of putative interaction were obtained. And complete sequences were cloned with PCR application from human testis epidermis cDNA library.
