摘要
研究了乳酸菌生长繁殖的环境条件(温度、接种量、起始pH等)和培养基组成(氮源、碳源、缓冲盐等),优化确定了乳酸菌发酵剂的适宜培养条件为:起始pH值为6.5,培养温度为37℃,培养基配比为麦芽糖∶乳糖(1∶1)2%、牛肉膏1.0%、缓冲盐A0.5%、NaCl0.25%、MgSO40.1%,接种量4%,进一步探索了半连续法进行高密度培养,结合优化的培养条件,可使乳酸菌的液体发酵活菌密度至1.1×1012CFU/mL。
This paper studied on the high cell density culture of Lactobacillus spp., and discussed the effects of cultural condition (temperature,inoculation capacity and initial pH) and media composition. The optimal culturing conditions were developed ,following:1% maltose, 1% lactose,1.0% beef extract,0.5% buffer salt A,0.25% sodium chloride, 0.1%magnesium sulfate, (pH6.5),4% inoculation capacity. Then high cell density culture was explored with one semi-continuous way and obtained the cells population over 1.1×10^(12)CFU/mL for 16 hours based on the optimal condition.
