脐带血造血干/祖细胞关键细胞周期蛋白D亚型的鉴定

Identification of the Crucial Cyclin D Isoform in Umbilical Cord Blood Hematopoietic Stem/Progenitor Cells

摘要 目的 确定控制脐带血造血干 /祖细胞增殖的细胞周期蛋白D (CyclinD)关键亚型。 方法 用Midi MACS免疫磁珠阳性分选的方法分离出脐带血CD34+ 细胞 ,在体外扩增以后 ,用RT PCR和WesternBlot分别在mR NA水平和蛋白水平检测CyclinD不同亚型的表达。 结果 mRNA在CyclinD1无表达 ,CyclinD2、CyclinD3有表达 ;蛋白在CyclinD1无表达 ,CyclinD2弱表达 ,CyclinD3高表达。 结论 控制脐带血造血干 /祖细胞增殖的CyclinD关键亚型是CyclinD3。 Objective To explore the isoforms of Cyclin D expressed in umbilical cord blood hematopoietic stem/progenitor cells. Methods The CD34 + cells were isolated from umbilical cord blood with immunomagnetic beads (Midi MACS). After expansion in vitro , the hematopoietic cells were harvested. The isoforms of Cyclin D were detected by RT PCR and Western blot from the mRNA and protein levels respectively. Results No Cyclin D1 mRNA expression was detectable, but Cyclin D2 and Cyclin D3 expressed at mRNA level. At protein level, the expression of Cyclin D1 protein was not detected, the expression of Cyclin D2 was weak and that of Cyclin D3 was strong.Conclusion Cyclin D3 isoform acts as the major role that determines the proliferation of umbilical cord blood hematopoietic cells.
