摘要
利用不同的电刺激条件对猪体外成熟卵母细胞进行孤雌激活试验 ,并探讨了不同成熟培养时间对孤雌发育的影响。试验结果 ,应用 2 .2 5kv/cm ,5 0 μs,1次电脉冲进行孤雌激活处理组获得了 2 2 % (2 2 / 10 0 )囊胚发育率 ,极显著高于 1.2 5kv/cm、5 0 μs、3次电脉冲处理组的 9.1% (9/ 98)和 2 .0 0kv/cm、5 0 μs、2次电脉冲处理组的 7.2 % (8/111) (P <0 .0 1) ;其囊胚细胞数为 6 7.8± 2 .7极显著 (P <0 .0 1)高于后两组的 5 3± 3.3、5 1.2± 5 .1。采用以上最佳电脉冲组处理成熟培养 4 2h、4 6h和 4 8h的 3组卵母细胞 ,各组间的卵裂率差异不显著 ,桑椹胚率 4 8h组为 5 7.8%(133/ 2 10 )极显著高于 4 2h组的 4 5 .7% (10 5 / 2 0 6 )和 4 6h组的 38.3% (88/ 2 11) (P <0 .0 1) ,4 2h组和 4 6h组间差异不显著 ;囊胚发育率 4 8h组 2 2 .6 % (5 2 / 2 10 )极显著高于 4 2h组的 9.6 % (2 2 / 2 0 6 )和 4 6h组的 6 .9% (16 / 2 15 ) (P<0 .0 1) ,4 2h组和 4 6h组间差异不显著 ;其囊胚细胞数 4 8h组为 6 8.9± 2 .8极显著高于 4 2h组的 5 5 .3± 3.4 (P <0 .0 1) ,显著高于 4 6h组的 5 0 .2± 5 .6 (P <0 .0 5 ) ,4 2h组极显著高于 4 6h组 (P <0 .0 1)。试验结果表明
In this experiment, it was designed to carry out parthenogenetic activation of swine oocytes derived from IVM under various situations of electric stimulus, discuss the effect of various length of mature culture time on parthenogenetic development. As a result, the blastocyst developmental rates of the group of oocytes under the parthenogenetic activation of once electric stimuli of 2.25 kv/cm for 50μs (22%,22/100) was significantly higher than that under the parthenogenetic activation of three times electric stimuli of 1.25 kv/cm for 50μs (9.1%,9/98) and that under the parthenogenetic activation of twice times electric stimuli of 2.00 kv/cm for 50μs(7.2%,8/111) (P<0.01) ,respectively; The cell number of blastocyst in the group of oocytes under the parthenogenetic activation of once electric stimuli of 2.25 kv/cm for 50μs (67.8±2.7) was significantly larger than these of the latter two groups(53±3.3)(P<0.01), (51.2±5.1)(P<0.05). Three groups of oocytes were respectively treated by the optimal electric stimuli mentioned above for 42h, 46h and 48h, there were no significant differences among the cleavage rates of the three groups, the morulae rate of the group of oocytes treated for 48h(57.8%,133/210) was significantly higher than that for 42h(45.7%, 105/206) and that for 46h(38.3%, 88/211)(P<0.01) respectively; there were no significant differences between the morulae rate of the group of oocytes treated for 42h and that for 46h.The blastocyst developmental rates of the group of oocytes treated for 48h(22.6%, 52/210) was significantly higher than that for 42h(9.6%,22/206) and that for 46h(6.9, 16/215)(P<0.01), there were no significant differences between that for 42h and that for 46h; the cell number of blastocyst in the group of oocytes treated for 48h(68.9±2.8) was significantly larger than that for 42h(55.3±3.4)(P<0.01) and that for 46h(50.2±5.6)(P<0.05) respectively; The cell number of blastocyst in the group of oocytes treated for 42h was significantly larger than that for 46h(P<0.01). The results of this experiment showed that different electric stimulus situations and different length of in vitro mature time significantly affected the blastocyst developmental rate and the cell number of blastocyst derived from swine oocytes treated by parthenogenetic activation.
