摘要
目的 :观察雌激素对去卵巢大鼠中脑黑质多巴胺能神经元的作用 ,探讨雌激素对帕金森病防治的可能性。方法 :对成年雌性大鼠行卵巢切除术 ,应用立体定向技术注射 6 羟基多巴胺 (6 OHDA)于大鼠中脑黑质 ,采用免疫组织化学的方法对黑质TH(酪氨酸羟化酶 )阳性神经元进行标记、计数 ;同时 ,对大鼠的行为学进行观测。结果 :在没有给予雌激素的去卵巢大鼠组 ,阿朴吗啡 (APO)诱导的旋转次数与其他对照组相比有显著性差异 (P <0 0 5 ) ,大鼠中脑黑质TH阳性神经元的数目与其他对照组比较也有显著性差异 (P <0 0 5 )。结论 :雌激素对雌性大鼠中脑黑质多巴胺能神经元具有保护作用 ,除了经过雌激素受体途径以外 ,还有其他的作用机制。
Objective:To investigate effects of estrogen on substantia nigral neurons in female rats with Parkinson disease which were ovariectomized and explore the possibility of estrogen to prevent and cure Parkinson disease.Methods:At 30 day after 6 OHDA injected stereotaxically into the right side of the mesencephic ventral tegmental area and substantia nigra pars sompacta of rats ovariectomized,we used immunohistochemical techniques to observe and count TH positive cells of the substantia nigra.In addition, we observed the behavior of rats at 30 day after injecting 6 OHDA.Last, we analysed results of various groups with statistical methods.Results:In the group without estrogen and the group with estrogen and tomoxifen the number of TH positive cells and the behavior of rats were significantly different compared with those in control groups ( P <0 05).Conclusion:Estrogen plays an important role in protecting the substantia nigral dopaminergic neuron of femal rats.
