摘要
提出了电渗析脱盐与单阴离子交换柱相耦合提取发酵液中谷氨酰胺的分离工艺。研究发现,将发酵液pH调节到谷氨酰胺等电点附近,通过电渗析将其中的无机盐脱除95%左右时,仍然能保证谷氨酰胺收率达到76%,电流效率为85%。根据谷氨酰胺和谷氨酸等电点差异,电渗析脱盐后的料液再通过单根阴离子树脂交换柱,可以去除其中的谷氨酸等杂质。该工艺大幅度地减少了树脂用量,有效地解决了谷氨酰胺在强碱和强酸环境中的转化问题,生产成本明显降低,谷氨酰胺的总提取收率达到60%左右,且产品纯度符合药典。
A new technique of one-step anion exchange for purification of glutamine from its fermentation broth was developed by using electrodialysis desalination to remove (NH4)2SO4 existing in the broth. Adjusting the pH of broth to the the isoelectric point of glutamine, most (NH4)2SO4 and only a little glutamine would transfer from the broth to the adjacent chamber through ion membrane during electrodialysis operation. When the desalination ratio reached 95%, the recovery ratio of glutamine and the current efficiency were 76% and 85%, respectively. After electrodialysis desalination, glutamine could be separated successfully from glutamic acid by passing the retained broth through a single anion exchange column. Experimental results show that this new process is effective in decreasing the amount of resins, preventing the decomposing of glutamine in strong acid and alkaline environment, and in the reduction of the production cost. The total yield of glutamine reaches about 60% and the purity of product satisfies the pharmaceutical standard.
