摘要
目的:观察放线菌酮对体外培养的黑素细胞的影响。方法:放线菌酮处理体外培养黑素细胞24h后,利用MTT法测定MC活力、流式细胞术测定MC凋亡率、电镜观察MC凋亡时形态学改变。结果:用40mg/l和80mg/l的放线菌酮分别作用24h后,黑素细胞活力明显降低,并出现典型的凋亡特征,染色质边集,核固缩,凋亡小体出现。流式细胞仪检测可见典型的凋亡峰。结论: 放线菌酮有抑制黑素细胞增殖和促黑素细胞凋亡作用。
Objective To inves tig ate the effects of cycloheximide on normal human melanocyte(NHM).Metho ds Melanocyte apoptos is was s tudied with MTT,and flow cytometry trans mitting electron micros copy.Res ults We added40mg/L and80mg/L cycloheximide in melanocytes s eparately,and the typital apoptos is was appeared24hours later,chromatin condens ation,nuelear pyknos is and apoptotic bodies formation.The res ults of cytometry s howed the typical apoptotic peak.Co nclus io n Cycloheximide could inhibit the proliferation of NHM and induce the apoptos is NHM.
