摘要
介绍了用斑点杂交技术,以Y染色体特异的3.5kb DNA片段为探针检测人血痕、人的组织碎块、牙齿、头发、羊水等的性别。样本经制备相当于0.1ul新鲜血的血痕,0.05 mg的组织块,带毛囊的头发1根,1/20颗牙齿牙髓质和2ul的羊水经本法检查,均能确定其性别。组织块经用福尔马林固定1周后,同样可以判断其性别,血痕性别的盲测结果全部正确。同时,对9种常见动物的血痕亦作了检查,均未显示阳性反应。本法准确灵敏,操作简单,值得进一步推广。
Results of sex determination of human bloodstains, tissue-debris, teeth, hairs, and amn-iotic fluids, by dot hybridization technique with Y chromosome specific 3.5 kb DNA probe, were reported. 0.1μl of fresh blood, 0.05mg of tissus-debris, one plucked hair, one 20th of a tooth pulp and 2μ1 of amniotic fluid were enough for sex determination by this method. The bloodstains stored longer than 4 years and the tissue-debris fixed by 10% formalin for one week could still be determined correctly. The blind test results of 6 bloodstains all were correct. These results showed that this method has many advantages, such as accuracy,sensitire, simple and fast so it is worthy to be spread in some laboratories.
