来自中间偃麦草基因组的类反转录转座子片段的克隆及其特征分析 被引量:5

摘要 中间偃麦草 [Thinopyrumintermedium ,(Host)BarkworthandDewey]是普通小麦 (TriticumaestivumL )遗传改良的重要基因源 ,已有许多重要基因导入普通小麦。本研究从中间偃麦草基因组克隆到一个类反转录转座子片段 ,命名为pTi2 8。该序列高丰度存在于中间偃麦草基因组 ,低丰度 (寡拷贝 )存在于普通小麦及其近缘种属硬粒小麦 (T durum)、黑麦 (Secalecereale)、小黑麦和簇毛麦 (Hynaldiavillosa)基因组 ,是中间偃麦草专化重复序列。序列对比分析结果显示 ,pTi2 8与大麦、小麦反转录转座子BARE 1和Wis 2 1A的同源性分别为 6 2 8%、70 8%。Southern及原位杂交结果表明 ,pTi2 8属散在型重复序列 。 Thinopyrum intermedium is an important source for improving the genetic variability of Triticum aestivum , and many useful agronomic traits have been introduced into T aestivum In this study, a retrotransposon like sequence was isolated from Th intermedium and designated as pTi28 This sequence was abundant in Th intermedium genome but rare in T aestivum genome and its relatives Sequence similarity analysis indicated that pTi28 was 70 8% similar to Wis 2 1A, a wheat retrotransposon like element and 62 8% identical to BARE 1 element from barley ( Hordeum vulgare ), respectively Southern blots and in situ hybridization results showed that pTi28 was an interspersed repeated DNA sequence and could a useful molecular marker for detection of Th intermedium chromatins in wheat background
