摘要
目的 :探讨光气是否诱导肺Ⅱ型细胞发生凋亡 .方法 :2 6只二级BALB/c小鼠 ,雄性 ,随机分为两组 :染毒组和正常对照组 (各 1 3只 ) .正常对照组小鼠以空气为对照 ,染毒组小鼠给予 1 1 .9mg/L剂量的光气 ,时间为 5min ,染毒后 4h ,比较两组小鼠肺脏的湿干比、病理学变化 .分离、培养小鼠原代肺Ⅱ型细胞并用电镜观察染毒组肺Ⅱ型细胞凋亡情况 .结果 :1 1 .9mg/L的光气染毒 5min ,小鼠肺脏湿干比 (6 .4 2± 1 .0 0 )比正常对照组 (4 .2 5± 0 .4 7)显著增加 (P <0 .0 5 ) ;光镜下观察肺组织可见肺水肿发生 ;单宁酸染色和电镜鉴定小鼠原代肺Ⅱ型细胞 ;电镜显示光气染毒肺水肿小鼠原代肺Ⅱ型细胞出现凋亡小体 .结论
AIM: To study apoptosis of alveolar type Ⅱ cells induced by phosgene in BALB/c mice. METHODS: 26 mice were divided into a negative group and a positive group with 13 in each. The mice in negative group were exposed to room air and the mice in positive group was exposed to 11.9 mg/L phosgene for 5 min. After 4 hours' exposure, all mice were immediately anesthetized. Lungs were analyzed for wet/dry weight ratio and pathological alternation. Mice alveolus type Ⅱ cells were isolated and cultured to observe its apoptosis under electron microscope after phosgene exposure. RESULTS: Lung wet/dry weight ratio was significantly higher in mice exposed to phosgene than in mice exposed to air ( P< 0.05). Phosgene caused pulmonary edema. Alveolus type Ⅱ cells were identified by tannic acid stainning and electron microscopy, and the apoptotic body in alveolus type Ⅱ cells were found in positive group. CONCLUSION: Phosgene may cause pulmonary edema and induce apoptosis of alveolus type Ⅱ cells in mice.
