摘要 目的 应用膜片钳全细胞记录技术 ,探讨ATP对坐骨神经损伤所引起的背根节神经元兴奋性的影响。方法 Wistar大鼠分为对照组和损伤组 ,损伤组建立大鼠坐骨神经离断模型 ,采用酶加机械分离方法急性分离背根节神经元 ,应用膜片钳全细胞记录技术测定神经元被动膜电特性 ,观察坐骨神经损伤后背根节神经元兴奋性的变化及ATP对其作用的影响。结果 与对照组比较 ,坐骨神经损伤后 1、10、10 0 μmol LATP引起神经元产生动作电位的去极化反应上升幅度比率分别为 45 8%、187%、75 %(P <0 0 1)。结论 坐骨神经损伤后背根节神经元兴奋性提高 。 Objective To investigate the effect of ATP on excitability of dorsal root ganglion neurons after sciatic nerve injury. Methods Rats were divided into control group and injury group. Rats in the injury group underwent transection of the sciatic nerves. Neurons were acutely dissociated from the dorsal root ganglion by a combination of mechanical and enzymatic method. Effect of ATP on excitability of dorsal root ganglion neurons was studied based on whole cell patch clamp after sciatic nerve injury. Results Rising frequencies of depolarization of action potentials evoked by 1, 10, and 100 μmol/L ATP were 458%, 187%, and 75%, respectively after sciatic nerve injury ( P <0 01). Conclusion After sciatic nerve injury, excitability of dorsal root ganglion neurons is increased, and reactive sensitivity of ATP receptor to ATP is enhanced.
