二氧化矽和石棉对小鼠肺泡巨噬细胞的细胞集落形成的影响

Effect of Colony-forming Capacity of Pulmonary Alveolar Macrophages from Mice Exposed to Silica and Asbestors

摘要 三种不同类型的粉尘,二氧化矽和青石棉分50μg 和200μg 两个剂量组,二氧化矽200μg 一个剂量组,给小鼠气管注入染尘。染尘后1,3和6个月,观察小鼠肺泡巨噬细胞体外集落形成能力的变化。结果表明:注入的二氧化矽和青石棉的量达50μg ,即可引起肺泡巨噬细胞体外集落形成能力的下降,细胞集落形成数及形成率均持续明显低于对照组。而在二氧化矽组则未观察到此现象。这说明二氧化矽和石棉对肺泡巨噬细胞增殖和分化具有细胞毒性。而惰性粉尘TiO_2对PAM 的增殖和分化则未构成不利影响。此外,我们发现GM-CSF 比M-CSF 更有利于小鼠肺泡巨噬细胞的集落生长,在前者作用下,集落形成率比后者约高1~2倍。 In order to observe colony-forming capacity(CFC)of pulmonary alveolarmacrophages(PAM)in vitro,groups of mice were intratracheally instilled with3 different kind of particles(Min-u-sil silica;crocidolite asbestos;TiO_2)atdose of 50μg and 200μg.The results showed that the total number of coloniesand the cloning efficiency of PAM in vitro were both significantly lower insilica and asbestos instilled animals than in the controls during the experimen-tal period from 1 to 6 months.The colony-forming ability of PAM fromTiO_2instilled animals was,however,almost copmparable to that of the con-trols.These results suggested that silica and asbestos have cytotoxicities on theproliferation and differentiation of PAM.In addition,it was found that bothGM-CSF and M-CSF have their ability to stimulate the growth of CFC ofPAM.But,the plating efficiency of CFC in GM-CSF is about 1-2 times high-er than in M-CSF.
