摘要
目的 观察急性一氧化碳中毒 (ACMP)后小鼠血小板膜糖蛋白 CD31、CD6 1和 CD6 2 p的变化及高压氧 (HBO)对其影响。方法 雄性昆明小鼠共 4 2只随机分成 7组 ,正常对照组、HBO(ACMP后 HBO治疗 )第 1、3、5天组及 ACMP第 1、3、5天组 ,每组各 6只。用流式细胞仪测定血小板膜上 CD31、CD6 1的平均荧光强度及 CD6 2 p的阳性率。结果 ACMP第 1天组和 HBO第 1天组 CD31的表达水平均明显高于正常对照组 (P<0 .0 1)。 ACMP第 1、3、5天组和 HBO第 1、3、5天组 CD6 1的表达水平均明显高于正常对照组 (P<0 .0 5或 P<0 .0 1) ;HBO第 3天组 CD6 1的表达水平明显低于同天 ACMP组 (P<0 .0 5 )。 ACMP第 1、3天组和 HBO第 3天组 CD6 2 p的表达水平均明显高于正常对照组 (P<0 .0 5 ) ;HBO第 1、3天组 CD6 2 p的表达水平均明显低于同天 ACMP组 (P<0 .0 5 )。结论 ACMP可显著增加CD31、CD6 1和 CD6 2 p在血小板膜上的表达 ,提示血小板活化反应增强 ;早期行 HBO治疗可显著减少CD6 1和 CD6 2 p的表达水平 ,而 CD31的表达水平无显著改变。
Objective To observe the changes of expression of platelet membrane glycoprotein CD31, CD61 and CD62p in mice after acute carbon monoxide poisoning and to observe the effect of hyperbaric oxygen on them. Methods 42 mice were randomly distributed into 7 different groups: normal control group, HBO 1st day ,3rd day,5th day after carbon monoxide poisoning groups;ACMP 1st day ,3rd day,5th day groups,The mean immunofluorescence intensity of CD31, CD61 or positive percentage of CD62p was measured by the flow cytometry (FCM). Results The expression of CD31 of the ACMP 1st day group and the HBO 1st day group was much higher than the normal control group(P<0.01). The expression of CD61 of each ACMP group and each HBO group was much higher than the normal control group (P<0.05 or P<0.01). Compared with the simultaneous ACMP group, the expression of CD61 of the HBO 3rd day group significantly decreased(P<0.05). The expression of CD62p of the ACMP 1st, 3rd day group and HBO 3rd day group was much higher than the normal control group (P<0.05). Compared with the simultaneous ACMP group, the expression of CD62p of the HBO 1st and 3rd day group both significantly decreased (P<0.05).Conclusions The expression of CD31, CD61 and CD62p on the platelet membrane significantly increased after acute carbon monoxide poisoning, indicating that platelet was activated. Early hyperbaric oxygen therapy could significantly decrease the expression of CD61 and CD62p on the platelet, while the expression of CD31 wasn't changed significantly.
