丙酸睾丸酮和神经节苷脂对交感神经化学损毁的影响

摘要 正常成年早小鼠♀经6-羟多巴胺(6—OHDA)15mg·kg^(-1)ip后24h颌下腺内去甲肾上腺素(NE)和多巴胺(DA)含量均下降95%以上,2wk时分别恢复到59和33%,丙酸睾丸酮(TP)给药后颌下腺内NE的含量提高63%,TP处理后再给6-OHDA,24 h和2 wk时腺体内的NE和DA含量仍在正常水平,但都低于正常动物TP处理后的水平;6-OHDA损伤早期给神经节苷脂(GM),24h颌下腺内NE和DA含量没有下降,而在2wk时有明显下降;TP处理后的动物在损伤早期再给GM,在2wk时NE和DA含量与正常动物TP处理后的水平相当,TP和GM在抗6-OHDA损伤中可能有协同作用。 The effects of testosterone propionate (TP) and the mixture of gangliosides (GM) on the sympathetic nerve lesion by 6-hydroxydopamine (6-OHDA) were investigated in the adult mouse submandibular glands (SMG). By HPLC techniques, it was demonstrated that the norepinephrine (NE) content in SMG increased 63% (as basic level) after the treatment with TP (1 mg·mouse-1, sc) on alternate days for 16 d. Single dose of 6-OHDA (15 mg·kg-1 ip) caused a decrease of NE and dopamine (DA) contents to less than 3% and 5% of normal control, respectively, 24 h after injection. The recovery of NE and DA levels went on slowly along with the course of regeneration of the lesioned sympathetic nerve terminals. The NE and DA contents reached 59% and 33%, respectively, 2 wk after lesion and fully recoverd by theend of the fourth week. If TP treatment was previously performed, NA and DA contents were still kept normal even 24 h and 2 wk after 6-OHDA, those in mice treated by TP only, but were higher than those of the lesioned control significantly. The TP and GM (20 mg·kg-1 ip) combining treatment of the mice caused a rise in NA and DA contents of SMG to basic levels at 2 wk of postlesion. The results indicated that TP had the action of promoting nerve regeneration and GM was also effective in protectiog the sympathetic terminals from 6-OHDA lesion. Their combination showed a certein potential action.
