摘要
本文主要用体外诱导不同T调节细胞的实验模型和单克隆抗体检测技术,分析白芍总甙(TGP)的双向免疫调节作用与T调节细胞的关系、TGP(0。40mg·L^(-1))可促进体外刀豆素A时间依赖性地诱生L_3T_4^+(T_(?)/i)细胞和Lvt—2^+(Ts/c)细胞.并分别拮抗环孢霉素A抑制Th细胞诱生和左旋咪唑抑制Ts细胞诱生的作用,提示TGP促进不同T淋巴细胞亚群诱生有明显的机能依赖性特征。实验还提示,TGP对小鼠脾淋巴细胞ConA增殖反应和体外抗体诱生呈现浓度依赖性双向作用.可能与其浓度依赖性改变L_3T_(?)^+/Lyt—2^+细胞比值有关。低浓度TGP促进ConA增殖反应和T细胞诱生可能是经激活巨噬细胞而实现的。
The percentages of L3T1+ (Th/i) cells which were selectively sup pressed by cyclosporin A (80 μg · mL-1) were increased obviously after the spleno-cytes of mouse were treated by con canavalin A (Con A, 2. 0 mg · L -1) plus total glucosides of paeony (TGP, 0.4mg · L-1)for 12 h. However, the percentages of Lyt - 2+(Ts/c) cells which were suppressed by levamisole (5. 0 mg · L-1) were enhanced significantly after the cells were incubated with ConA (2. 0 mg · L-1) plus TGP(0.4mg · L-1) for 40h. It was found that Con A induced proliferations and the anti - SRBC antibodies (IgM) production by mouse splenocytes were enhanced by invitro treatment with lower concentrations of TGP.but diminished with higher concentrations. At lower concentrations of (0.2mg · L-1) of TGP. the percentages of L3T1+ cells and the ratios of L3T1+/Lyt - 2+ cells were increased , at higher concentration ( 1. 6mg · L-1)of TGP, the percentages of Lyt -2+ cells were augmented and the ratios of L3T1+Lyt - 2+ cells dropped down. These might be related to its dual-effects. The results also showed that ConA induced proliferative responses of mouse splenocytes and the induction of Th cells were increased by lower concentrations of TGP via the activations of macroph ages. In conclusion, these data suggested that TGP could promote the activation of macrophages and the induction of different T lymphocyte subsets in vitro, depending on different stages of activation of the cells and concentrations of TGP treated, which might account for the immuno - regulatory effects of TGP.
