摘要 报道了重组人白细胞介素-2(rHuIL-2)高表达菌株的最佳培养和表达条件、表达产物的分离纯化、复性条件对rHuIl-2活力的影响及高纯度rHuIL-2的性质鉴定。利用制备性SDS-聚丙烯酰胺凝胶电泳(PAGE)进行rHuIL-2的纯化,回收率可达80%以上,纯度达95%以上,其比活为1~2×10~6 U/mg,能在体外维持CTLL和CTC细胞的增殖,其分子量、等电点、氨基酸组成、N-末端分析结果与德国Boehringer公司的同类产品基本一致。 The most optimal conditions of fermentation and expression of the strain of producing highly rHulL-2 activity, its isolation and purification, and effect of renaturation conditions on its activity were presented. The purification was performed by preparative SDS-PAGE. The recovery, purity and specific activity of the purified rHuIL-2 were 80%, 95% and 1~2×10~6 U/rag respectively. The purified rHuIL-2 could maintain CTLL and CTC proliferation in vitro, and the data of purified rHuIL-2 were in agreement with that of the preparation from Boehringer Manheim in the aspects of molecular weight, isoelectric point, amino acid composition and N-terminal.
