本实验观察到活性氧自由基(光照虎红酸钠,RB),在低浓度(RB:10^(-6)M)时,有增加豚鼠乳头肌收缩幅度和出现自发收缩的效应;在高浓度时(RB:10^(-5)~10^(-4)M),显著地增加乳头肌静息张力,以致出现“挛缩”。设想这可能与细胞损伤及胞内钙离子异常有关。
It is known that photoactivation of rose bengal (RB) can produce activated oxygen intermediates——mainly singlet oxygen and small amount of hydroxyl free radical.We observed that illuminationof low concentration of RB (10^(-6)M)in the bath could enhance contraction amplitude of papillary muscle of guinea pig heart by 248.7% of the control and evoke spontaneous contractions. Resting tension of papillary muscle showed significant increase(eventually approaching 'rigor'). No contraction was found when concentration of RB in the bath reached 10^(-5) M-10^(-4) M and even after 30min washing. It is thus speculated that myocardial injury might be initiated directly by activated oxygen intermediates and related to the increase of intracellular calcium load.
