新辅助化疗对乳腺浸润性导管癌端粒酶活性的影响 被引量:1

摘要 目的 :探讨新辅助化疗对乳腺癌细胞端粒酶活性的影响。 方法 :2 9例原发性乳腺浸润性导管癌女性患者 ,术前细针穿刺行细胞学诊断 ,诊断明确后行CEF(环磷酰胺、表阿霉素、氟脲嘧啶 )方案化疗 ,化疗后 3~ 5天行手术治疗。术前细针穿刺留置标本及术中所取新鲜癌组织 ,均采用TRAP ELISA法定量检测端粒酶活性。 结果 :化疗后乳腺癌组织端粒酶活性较化疗前显著降低 (0 .771± 0 .4 4 2vs 1 .1 72± 0 .5 0 1 ,P <0 .0 1 ) ,化疗前T1、T2、T3期肿瘤端粒酶活性分别为 0 .6 2 6± 0 .1 4 0、1 .0 2 9± 0 .4 1 0、1 .5 1 6± 0 .4 6 1 ,治疗后分别为 0 .2 30± 0 .0 5 5、0 .6 2 0±0 .2 80、1 .1 2 4± 0 .4 2 3(P <0 .0 5 )。 结论 :新辅助化疗显著降低乳腺癌组织端粒酶活性 ,随着肿瘤体积增大 ,化疗药物对端粒酶活性的抑制作用减弱。 Objective:To investigate the significance of telomerase activity in infiltrating ductal carcinoma with respect to neoadjuvant chemotherapy effect. Methods:Fine needle aspiration (FNA) specimens were obtained from twenty nine patients with primary breast cancer pre neoadjuvant chemotherapy.All patients were given neoadjuvant chemotherapy(CEF regimen, Cyclophosphamide 500 mg/m 2 ; Epirubicin 50~60 mg/m 2 , Fluorouracil 500 mg/m 2 ) and were performed operation on post chemotherapy 3th day to 5th day,cytology and telomerase activity of specimens were analyzed. The telomerase activity was measured Telomerase PCR ELISA based on a polymerase chain reaction (PCR) assay known as TRAP (telomeric repeat and amplification protocol) assay. Results:The optic density (OD) of telomerase activityis is 1.172±0.501 pre chemotherapy whereas post chemotherapy that is 0.771 ±0.442 ( P < 0.01). The telomerase activity in T1, T2 , T3 is 0.626±0.140, 1.029±0.410, 1.516 ±0.461, respectively pre chemotherapy while that is 0.230±0.055, 0.620±0.280, 1.124± 0.423 post chemotherapy( P <0.05, pre chemotherapy vs post chemotherapy) with a significant reduction of decreasement among the groups. Conclusion:Neoadjuvant chemotherapy decreased telomerase activity of breast cancer.
