摘要
国外已建立了4h水解酶色原底物系列,成功地用于厌氧菌的分类与鉴定,鉴于国内的无系列水解底物,我们对厌氧菌系列脱氢酶进行了研究,以便建立一种快速鉴定系列。为此选择类杆菌的参考株作为分类单位,以不同的糖、氨基酸和有机酸的天然底物纸片与细菌完整细胞进行酶学反应,并以四唑盐受氢后以变色效应来指示酶的存在。筛选了18项脱氢底物与色氨酸、尿素一起,组成20项以检测细菌脱氢酶为主的系列,研究发现完整的菌细胞可以较好地表现底物的脱氢酶活性,酶的分布具有菌种特征,这个系列在有氧条件下,30~60min出现明显的可见反应。以4株脆类杆菌参考株进行24次重复试验,其中9项的阳性概率为0.99,7项为0.01,其它4项分别是0.03,0.125,0.5和0.66。尤其胆酸脱氢酶与常规胆汁抑制试验有较好的相关性,有可能取代厌氧菌分类中有要意义的20%胆汁抑制实验。
The chromatic substrates systems of hydrolase have been used to classify and identify anaerobic bacteria abroad. Since there are not any the substrates at home, the dehydrogenase system of anaerobes has been set up for rapid identifying the bacteria. In this system, the nature substrates such as sugar, amino acid and organic were used to react with the constitutive dehydrogenases of bacteroides and the reduced NAD^+ transfered the released hydrogen proton to TTC which then changed to red colour that indicates the enzymes activity. The system consisted of 18 kinds of dehydrogenic substrates, indole and urea. The result proved that the enzymatic reactions were characteristic for species of the bacteria and completed after incubation in air for only 30-60 minutes. 4 strains of bacteroides fragilis were tested 24 times, the positive probability were evaluted, 0.99 was in 9 of the system, 0.01 was in 7, 0.08, 0.125, 0.5 and 0.66 was in the others. Especially, bile acid dehydrogenic reaction is related with the 20% bile fluid inhibitive test that is a important marker in taxonomy of bacteroides.
