摘要
目的 :建立同时测定莲子心中酚性生物碱 :莲心碱、异莲心碱、甲基莲心碱含量的RP -HPLC法以及酚性生物碱的特征图谱。方法 :采用RP -HPLC法 ,流动相为乙腈 水 三乙胺 (2 5∶75∶0 .1 ,用磷酸调 pH3.3) ,紫外检测波长 2 82nm ,流速0 .8mL·min-1 ,进样量 2 0 μL。结果 :莲心碱、异莲心碱、甲基莲心碱线性方程显示线性关系良好。特征图谱研究确定了酚性生物碱 1 6个共有峰。结论 :本方法简便、准确 ,重现性好 ,专属性强 ,可为莲子心中生物碱类物质标准提取物质量控制提供参考。
OBJECTIVE To develop an RP HPLC method for the quantitative analysis of liensinine?isoliensinine and neferine in Plumula Nelumubinis simultaneously and establish a characteristic chromatogram. METHODS The analysis was conducted by RP HPLC,CH 3CN H 2O Triethylamine (adjust pH 3.3 with H 3PO 4)=25∶75∶ 0.1 as mobile phase and UV detection at 282 nm .Flow rate was 0.8 mL ·min -1 and 20 μL was injected.RESULTS The linear relations of liensinine?isoliensinine and neferine were very good. In the chromatogram,16 common peaks of phenolated alkaloids were confirmed. CONCLUSIONS The method is simple and accurate with a good reproducibility and specificity and can be used as a quality control method for preparations of standard extract of the alkaloid in PLumula Nelumbinis .
