摘要
为研究大鼠急性心肌梗死 (AMI)后梗死周围区左室心肌Kv4 .2mRNA表达量的变化和美托洛尔、缬沙坦对其影响 ,将 4 1只SD雌性大鼠随机分为 2组 ,每组再随机分为对照组、手术组、美托洛尔组、缬沙坦组等四个亚组 ,应用RT PCR方法检测大鼠AMI后 7天 ,30天梗死周围区域左室心肌Kv4 .2mRNA的水平 ,并以图像分析系统对其进行半定量分析。结果 :分别在 7天 ,30天 ,与对照组相比 ,手术组、美托洛尔组与缬沙坦组Kv4 .2mRNA水平均明显降低 (P <0 .0 0 1)。与手术组相比 ,美托洛尔组第 7天Kv4 .2mRNA水平明显升高 (78.4 5± 1.72vs 6 7.2 9± 4 .6 4 ,P <0 .0 5 ) ,第 30天反而降低 (77.4 1± 2 .4 7vs 83.4 0± 2 .86 ,P <0 .0 5 ) ;而缬沙坦组第 7,30天Kv4 .2mRNA水平均明显高于手术组 (79.89± 6 .4 3vs 6 7.2 9± 4 .6 4 ,P <0 .0 5 ;95 .0 7± 2 .30vs 83.4 0± 2 .86 ,P <0 .0 5 )。第 7天 ,美托洛尔组和缬沙坦组相比无差异 ,而 30天后者较高 (95 .0 7± 2 .30vs 77.4 1± 2 .4 7,P <0 .0 1)。与第 7天相比 ,30天时手术组和缬沙坦组Kv4 .2mRNA水平均有增高 ,而美托洛尔组没有变化。结论 :与美托洛尔相比 ,缬沙坦似乎能更大程度的提高大鼠心肌梗死后Kv4 .2mRNA水平。
To investigate the changes of the mRNA expression of Kv4.2 potassium channel after acute myocardial infarction(AMI) in SD female rat, and the effects of the drug, metoprolol or valsartan, female SD rats(n=41) was randomly separated into two groups(n=20, 21), which were randomly separated into four subgroups, post-MI group(n=5, 6), sham group(n=5, 5), metoprolol group(n=5, 5) and valsartan group(n=5, 5), respectively. The level of gene transcription of Kv4.2 potassium channel was examined by semi-quantitative RT-PCR. Semi-quantitative analysis of Kv4.2 gene transcription was performed by pathological image analytic system. Results: Compared with sham group, the mRNA expression of Kv4.2 was significantly decreased in post-MI group after 7, 30days(67.29±4.64 vs 98.88±6.96, P<0.001;83.40±2.86 vs 94.68±4.26 , P<0.001), which could be changed by metoprolol or valsartan. Metoprolol could improved the gene expression of Kv4.2 in rats with AMI after 7 days(78.45±1.72 vs 67.29±4.64 ,P<0.05), however, decreased that in rats with AMI after 30 days(77.41±2.47 vs 83.40±2.86,P<0.05). Differed from metoprolol, valsartan could improved the gene expression of Kv4.2 in rats with AMI not only after 7 days(79.89±6.43 vs 67.29±4.64,P<0.05), but also after 30 days(95.07±2.30 vs 83.40±2.86 ,P<0.05). The mRNA expression of Kv4.2 was notably increased to a higher level in post-MI group and valsartan group after 30 days than 7 days(83.40±2.86 vs 67.29±4.64, P<0.001; 95.07±2.30 vs 79.89±6.43, P<0.01, respectively). But no difference of Kv4.2 expression was found in metoprolol groups between 7 day and 30day. Conclusions: Compare with metoprolol, Valsartan is likely to increase the higher level of Kv4.2 mRNA in rat after AMI.
