摘要
目的 :探讨剔毒护肝方及其组成药物黄芪、莪术、叶下珠对人肝癌细胞增殖及端粒酶活性的影响。方法 :剔毒护肝方 (1g/ml) ,黄芪 (0 3 g/ml) ,莪术 (0 3g/ml) ,叶下珠 (0 4g/ml) ,分别制成水煎剂 ,按大鼠体重 10mg/kg ,灌喂正常大鼠 ,2次 /d ,连续 3天 ,于第 4天 ,1次给予全日剂量后 1小时采血 ,制备药物血清。以正常鼠血清为阴性对照 ,将药物血清温育Bel 740 2人肝癌细胞。噻唑蓝 (MTT)比色法测定细胞增殖 ,多聚酶链反应 酶联免疫吸附法 (PCR ELISA法 )检测细胞端粒酶活性。结果 :剔毒护肝方和莪术均可抑制Bel 740 2细胞的增殖 (P <0 0 1) ,黄芪、叶下珠不能抑制Bel 740 2细胞的增殖 (P >0 0 5 )。剔毒护肝方和莪术还可抑制Bel 740 2细胞端粒酶活性 (P <0 0 1)。结论 :剔毒护肝方和莪术均可抑制Bel 740 2细胞的增殖 ,其部分机制可能在于抑制Bel 740 2细胞端粒酶活性。剔毒护肝方对Bel 740 2细胞增殖及端粒酶活性的抑制作用主要是来自于方中莪术的药理作用。
Objective: To investigate the influence of Tidu Hugan recipe and separated recipes(astragalus, zedoary and phyllanthus)on the proliferation and telomerase activity of human hepatocarcinoma cell. Methods: Tidu Hugan recipe, astragalus, zedoary and phyllanthus were prepared as decotion respectively, and fed to the normal rats. The drug sera were collected from the rats and incubated with cultured Bel-7402 human hepatocarcinoma cells. While the normal rat sera were used as the control drug. Later, observed the proliferation of Bel-7402 cell by MTT colorimetric assy, tested the telomerase activity of Bel-7402 cell by using PCR-ELISA method. Results: Both Tidu Hugan recipe and zedoary not only can inhibit the proliferation of Bel-7402cell( P <0.01),but also can suppress the telomerase activity of Bel-7402 cell( P <0.01). Astragalus and phyllanthus can not inhibit the proliferation of Bel-7402 cell( P >0.05). Conclusion: Both Tidu Hugan recipe and zedoary can inhibit the proliferation of Bel-7402 cell. The main mechanism may be they can suppress the telomerase activity of Bel-7402 cell. The influence of Tidu Hugan recipe on the proliferation and telomerase activity of Bel-7402 human hepatocarcinoma cell are result from the pharmaceutical function of zedoary.
