摘要
构建人组织型纤溶酶原激活剂 (t PA)与绿色荧光蛋白 (GFP)融合表达的植物表达载体pCA130 2NSF tPA。将重组载体转入根瘤农杆菌GV310 1中 ,通过叶圆盘法转化烟草。转化植株经PCR鉴定整合有t PA基因 ,通过荧光显微镜可检测到绿色荧光蛋白 (GFP)的表达。
Fusion gene of human tissue-type plasminogen activator(t-PA) and green fluorescent protein(GFP) was inserted into pCAMBI1302 for constructing the plant expressing vector pCA1302NSF-tPA.Then the recombinant vector was transformed into A.tumefaciens GV3101 and the fusion gene was transformed into Nicotina tobaccum L.W38 line with the help of GV3101.Transgenic tobacco plants were detected by PCR method and the result shows that t-PA cDNA was integrated into the genome of tobacco.The recombination t-PA-GFP could be detected through fluorescent telescope in leaf and its protoplasts. So a effective and rapid way to screen positive transgenic tobacco plants was established.
