肾综合征出血热病毒感染促进人内皮细胞与PHA或TNF—α刺激白细胞的粘附 被引量:1

摘要 应用胶原酶消化法获取人脐带静脉内皮细胞,在体外传代培养.以 HFRS 病毒感染培养的人脐带静脉内皮细胞做为模型,采用刚果红染色法在感染后的12小时、3天、5天分别检测与 PHA 活化淋巴细胞及 TNF—α刺激嗜中性粒细胞的粘附作用.发现在第3天、第5天感染内皮细胞与上述两种细胞的粘附作用明显高于未感染的内皮细胞,提示 HFRS 病毒的感染可能促进了人脐带静脉内皮细胞粘附分子的表达. Human umbilical vein endothelial cells(HUVE)were obtained by collagenase digestion and passage in vit-ro.The cultured HUVE cellswere infected with hemorrhagic fever with renal syndrome virus.Twelve hours,3and 5 days after infection,the adhesion of the endothelial cells with PHA activated lymphocytes or TNF—αstimulated neutrophils was detected by rose bengal staining.It was found that 3 and 5 days after infection theadhesion of infected endothelial cells with the two kinds of cells above markedly incresed than that of uninfectedendothelial cells.It is suggested that infection of HUVE cells with hemorrhagic fever with renal syndrome virusmay increase their expres sion of adhesion molecules.
