摘要
穿孔素是一种由杀伤细胞(NK 细胞和 CTL 细胞)产生的具有杀伤功能的蛋白质,其在 SLE发病中的作用文献中尚未见报道.我们探索性地采用斑点杂交和 Northern 印迹杂交的方法,检测了 SLE 活动期和非活动期患者外周血单个核细胞的穿孔素 mRNA 量.结果显示:活动期 SLE 患者穿孔素 mRNA 增多(9.25±0.50,n=11),而非活动期 SLE 患者(2.50±1.47,n=10)在正常人对照范围内(1.61±1.59,n=11).活动期与非活动期及正常人对照间有非常显著的差异(P<0.01).Northern 印迹杂交提示活动期患者的穿孔素 mRNA 具有正常形态(大小约为3.0Kb).LDH 释放法测定 NK 细胞杀伤活性表明,活动期患者(16.2±8.1%)与非活动期患者(17.2±7.5%)均低于正常对照(39.6±15.5%),P 值均小于0.01.活动期患者穿孔素 mRNA 升高,而 NK 活性降低,两者不相关,其原因有待进一步研究.但穿孔素 mRNA增高与血清中抗 dsDNA 抗体水平增高相关(γ=0.76,P<0.01).实验结果提示,穿孔素 mRNA 表达水平或许可以作为 SLE 患者病情活动性的一个诊断指标.
Since abnormality of NK cell activity has been found in some systemic Lupus Erythematosus (SLE)patients and perforin released by NK cells is one factor which mediates the cytotoxic effect of the cells on targets,the expression of perforin gene in PBMNC from SLE patients has been investigated here on mRNA level using dot blot and northern blot hybridizations.Dot blot data showed that the amount of perforin mRNA in active SLE patients(9.25±0.50,n=11)was signficantly higher than that in nonactive SLE patients(2.50±1.47,n=10)(p<0.01)and that in normal control(1.61±1.59,n=11)(P<0.01).Northern blot hybridization analysis further confirmed the increase of perforin mRNA in active SLE patients and showed that the size of perforin mRNA was in normal range(about 3.0Kb).However,NK cell activty was signficantiy decreased in both active(16.2±8.1%,n=11)and nonactive(17.2±7.5%,n=10)patients(normal controls:39.6±15.5%,n=11)(P<0.01).The discrepancy between the perforin mRNA and NK activity in SLE patients needs to be investigated.Interestingly,there was a correlation between high perforin mRNA and high level of anti-dsDNA antibody in active SLE patients(r=7.6,P<0.01),suggesting that high perforin 8ene transcription might be a new parameter indicating the activation of the disease.
