摘要
采用硫酸铵分段盐析、DEAE-Sepharose CL-6B 离子交换层析、Sephadex G-75凝胶过滤、聚焦层析和 HPLC 五个步骤从精液中提纯了 PSA,并制备了 PSA 抗血清。提纯的 PSA 分子量33000,等电点6.75-6.91,SDS—PAGE 呈单一带。在免疫双扩散中与进口抗 PSA 抗体和自制抗 PSA 血清形成的沉淀线互相融合,提纯的 PSA 与自制的抗 PSA 血清作免疫电泳鉴定,为单一沉淀线。
Prostate specific antigen(PSA)was isolated from human seminal plasma by the steps of fractional precipitation by ammonium sulfate(30-70% saturation),DEAE-sepharose CL-6B anion exchang chromatography,gel filtration on Sephadex G-75,chromatofocusing and HPLC.Rabbit anti-PSA serum was prepared furthermore.The molecular weight of purified PSA is 33 000 and pi is between 6.75 and 6.79.The single band showed in SDS-PAGE.The fuset immunoprecipitin line formad in double immunodiffusion of PSA reacting with imported PSA antibody and prepared anti-PSA serum.In immunoelectrophoresis,only one precipitin line was abtained between anti-PSA serum and purified PSA of seminal plasma.Neither purified PSA reacted with sheep anti-human serum,nor anti-PSA serum do with normal human serum.
