摘要 目的:建立高效液相色谱法测定核黄素四丁酸酯的含量和有关物质。方法:采用迪马公司Diamonsil c18色谱柱(250 mm×4.6 mm,5μm),以甲醇-水(80:20)为流动相,流速为1.0 mL·min-1,用外标法测定,紫外检测波长为447nm。结果:在0.24-1.2μg范围内,色谱峰面积与浓度呈良好的线性关系,r=0.9999;回收率为100.8%(RSD=0.48%,n=5),最低检测量为4ng。结论:该法准确、简便、灵敏,可用于核黄素四丁酸酯的含量测定和有关物质检查。 Objective: To establish a HPLC method for the determination of riboflavine tetrabutyrate and its related substances. Methods:The Diamonsil C18(250 mm×4. 6 mm,5μm)column was used,the mobile phase was metha nol-water (80:20) ,the flow rate was 1.0 mL·min -1 ,the detection was carried out with an ultraviolet detector at 447 nm. Riboflavine tetrabutyrate was measured with the external standard method. Results:The calibration curve was linear in the range of 0.24-1.2μg,r=0.9999. The average recovery was 100.8% (RSD=0.48%, n=5). The minimum detection amount of riboflavine tetrabutyrate was 4 ng. Conclusion: This method is accurate, simple and sensitive, and it can be used for the assay or related substances control of riboflavine tetrabutyrate.
