低氧诱导因子-1α表达上调在大鼠局灶性脑缺血耐受中的意义 被引量:3

摘要 目的:观察局灶脑缺血预处理对低氧诱导因子-1α(hypoxia-inducible factor-1α,HIF-1α)表达的影响,探讨其在脑缺血耐受中的意义。方法:SD大鼠随机分为预缺血组、假手术组及对照组,前两组分别在2h大脑中动脉缺血(MCAO)前3d给予10min的预缺血或假手术,MCAO后24h处死,对照组给予两次相隔3d的假手术,比较各组梗死体积及HIF-1α的表达。结果:对照组未见HIF-1α表达,预缺血组HIF-1α表达高于假手术组,梗死体积较假手术组减少53.15%(P<0.01)(P<0.01)。结论:病灶性脑缺血可诱导HIF-1α表达。HIF-1α表达上调可能是脑缺备耐受产生的分子机制之一。 Objective: To investigate the expression of hypoxia-inducible factoi-1α( HIF-1α) in ischemic toIerance induced by focal ischemic preconditioning in rals. Methods:32 rats were divided into 3 groups in which control group received sham sur-gery (SS) only, and the other two groups received 2h middle cerebral artery occlusion ( MCAO) followed by 22h reperfusion with 10min ischemic preconditoning or SS 3d before. Infarct volume and expression of HIF-1α were evalualed in each group Results: Compared with the SS group, there was a smaller infarct volume and stronger HTF-1α immunoreactivity in PC group. Conclusions: 10min MCAO was strong enough to induce ischmic tolerance and increase HIF-1α expression. Up-regulation of HIF-1α might be a key event in [he molecular mechanism of focal cerebral ischemic tolerance.
