摘要
雄性Wistar大鼠分别经口染毒黄磷1.5mg/kg,亚砷酸钠20mg/kg、四氯化碳500mg/kg,每周3次,持续12周;采用聚丙烯酰胺凝胶电泳分离,软镭射光密度扫描仪定量测定血清乳酸脱氢酶(LDH)同工酶,酶组化染色,微机图象处理光密度定量分析肝组织5'核苷酸酶LDH。结果表明,血清LDH_5在3种毒物中毒性肝损害过程中都明显升高且与其肝脏病理和酶组织化学改变相吻合,能敏感地反映肝细胞实质性损害及其程度,特别是反映黄磷,亚砷酸钠的肝损害比谷氨酸丙酮酸转氨酶敏感、可靠。
Male Wistar rats were treated orally yellow phosphorus 1.5 mg/kg, sodium arsenite 20 mg/ kg, carbon terachloride 500 mg/kg, 3 times a week up to 12 weeks,LDH isoenzymes in serum were separated by polyacrylamidegel electrophoresis and measured by Soft Laser Scanning Denstometer; liver LDH, 5′ nucleotidase were measured by morphometry in histocytochemistry. In rats of three groups,Serum LDH_5 were increased during toxic hepatic injury, and such enzymological changes were consistent with liver pathological and histocytochemical changes. Elevated serum level of LDH_5 is reliable reflection of parenchymal hepatic injury and its extent. Serum LDH_5 is more sensitive and stable than total LDH, alanine aminotransferase in detection of hepatic damage induced by phosphorus and sodium arsenite.
