摘要
为了确立台湾青枣组织培养的消毒方法 ,研究了多菌灵、酒精、升汞和吐温 2 0这 4种消毒剂对台湾青枣组织培养的消毒效果和对其愈伤组织生长的影响。结果表明 :提前 5d喷 6 0 0倍液多菌灵 ,能明显减少污染率 ;经 30s75 %酒精消毒后 ,再用 0 1%升汞加 3～ 5滴吐温 2 0消毒 5min ,既能减少污染 ,又能促进外植体愈伤组织生长。
Effects of sterilization on explants and of growth on callus were studied in the tissue culture of Taiwan green jujube of Zizyphus mauritiana by means of 1/600 carbendazim, 75%alcohol, 0.1% mercuric chloride and Tween-20 in order to establish the sterilization method of the tissue culture. The results were as follows: The explant pollution rate was decreased by spraying 1/600 carbendazim 5 days before the explants were used. 75% alcohol sterilized the explants for 30 seconds, 0.1% mercuric chloride with 3～5 drops of Tween-20 sterilized the explants for 5 minutes, which not only decreased the explant pollution rate, but also promoted the callus growth.
