摘要
目的:观察甘油三酯对体外培养大鼠胰岛细胞分泌功能及对细胞内钙离子平均荧光强度的影响。方法:体外分离SD大鼠胰岛细胞进行单层细胞培养,与不同浓度甘油三酯(0、0.25、2.5、5.0、10.0mmol/L)共同孵育72小时,进行葡萄糖负荷试验,放射免疫法检测培养液中胰岛素水平,以评价胰岛细胞功能变化;采用Flou-3/AM荧光染色法,共聚焦显微镜下观察细胞内钙离子荧光强度的强弱,以间接反映细胞内游离钙的水平。结果:5.0、10.0mmol/L甘油三酯可抑制胰岛细胞的高糖刺激下的胰岛素分泌(p<0.05);甘油三酯组(2.5、5.0、10.0mmol/l)细胞内钙离子平均荧光强度均明显增强,与对照组比较有显著性差异(p<0.05)。结论:甘油三酯可以导致胰岛细胞内游离钙水平异常升高,影响细胞的分泌功能。
Objective: To study the effect of triglycerides (TGs) on function and intracellular free calcium of rat islet cells in vitro. Methods: Islet cells of SD rat were separated for monolayer cell culture and were treated with different concentrations of TGs(0、0.5、2.5、5.0、10.0 mmol/L intra-lipid) for 72 h. Insulin level in the cell culture media was measured after glucose load test. Meanwhile the Flou-3/AM was applied to probe intracellular free calcium concentration and the fluorescent intensity was detected using laser confocal microscopy technique. Results: High glucose (27.8 mmol/L)-induced insulin secretion was inhibited by 5.0 mmol/L TGs or 10.0 mmol/L TGs; fluorescent intensity in TGs groups(2、5.0、10.0 mmol/l) was significantly higher than that of control group. Conclusion: TGs might induce intracellular free calcium elevation in intro islet cells and have a inhibitory effects on the cell function.
