二氢青蒿素对大鼠系膜细胞增生的抑制作用 被引量:1

摘要 目的 :观察二氢青蒿素 (DHA)对大鼠系膜细胞 (MC)增生的抑制作用 .方法 :采用MTT掺入方法 ,观察不同浓度DHA和不同作用时间各组体外培养的MC增生水平 .结果 :DHA对MC增殖有显著的抑制作用 ,其抑制作用呈浓度依赖性和时间依赖性 .LD50 为 19 7μmol/L .DHA作用 72h后 ,从 0到 10 0 μmol/L浓度梯度 ,以10 0 μmol/L对MC增生的抑制作用最为显著 ;DHA 30 μmol/L作用 0~ 72h ,以 72h对MC增生的抑制作用最为显著 (P <0 0 1) .结论 Objective: To investigate the inhibiting effect of dihydroartemisinin on proliferation of rat mesangial cells in vitro. Methods: Rat mesangial cell line was cultured in vitro, incubating with different concentrations and different time of DHA. Cell proliferation was assessed by MTT colorimetric assay. Results: DHA could significantly inhibit the proliferation of mesangial cells, with a relatively low LD 50, 19.7?μmol/L. The inhibiting effect shows a dose-dependent and time-dependent manner. Inhibition at 100?μmol/L of DHA is the most significant among the different concentrations of DHA (0?μmol/L to 100?μmol/L). And mesangial cells are the most significantly inhibited after 72 hours of incutation with DHA, among the different time courses (0?hour to 72?hours) (P<0.01). Conclusion: Dihydroartemisinin could significantly inhibit mesangial cells proliferation.
