兔骨髓基质细胞体外培养的生物学特性 被引量:1

摘要 目的 研究兔骨髓基质细胞 (MSC)体外培养状态下的生物学生长特性 ,为骨 /软骨组织工程提供实验理论和技术支持。 方法  抽取兔骨髓分离MSC ,连续培养 5代。描绘 1~ 5代MSC生长曲线、计算倍增时间 ,测定细胞贴壁率和细胞活性。 结果  第 1~ 3代传代细胞生长曲线基本相同 ,倍增时间 2 0h ,贴壁率无明显差异 ,接种后 12h贴壁率为 96 % ,成活率大致相等 ,为 95 % ;第 4、5代细胞倍增时间延长 ,贴壁能力及细胞活力下降。 结论 原代及第 1~ 3代MSC体外培养生长稳定 ,增殖迅速 ,贴壁率高 ,活力良好 ,是骨 Objective To investigate the biological characteristics of rabbit marrow stromal cells(MSC)in vitro and supply bone /cartilage tissue engineering with fundament theory and technologic support.Methods Rabbit bone marrow was harvested from tibias and iliac crests.MSC were separated by centrifuging and cultured in the DMEM medium without any induced factors.The confluent MSC were released and consequently subcultured for 5 passages.The following dates were contiuously measured:growth curve,cell doubling time,confluent rate,cell activity.Results The growth curves and the doubling times of passage 1~3 were similar,and the doubling time of passage 2 was 20h.Both them began decreasing or prolonging from passage 4 gradully.So do the cell activity and confluent rates of passage 4and 5.Conclusion MSC of passage 1~3 proliferated rapidly and grew stably in vitro.The MSC of passage 1~3 may be well used in autogeneous bone/cartilage tissue engineering.
