牧草植物鹰嘴紫云英的遗传转化

摘要 鹰嘴紫云英(Astragalus cicer)是一种优良的豆科牧草。 15kD玉米醇溶蛋白是一种富硫的蛋白质,含硫氨基酸占总氨基酸量的15.63%。本文通过农杆菌(Agrobacterium)的介导将rbcS启动子调控的15kD玉米醇溶蛋白基因的嵌合质粒导入鹰嘴紫云英,得到转化的植株。这些植株具有NPTⅡ酶活性,抗卡那霉素。Southern blot分子杂交表明,15kD玉米醇溶五白基因已转化并整合入鹰嘴紫云英的核基因组中。 Astragalus cicer is a fine legume forage.15kD zein is a sulphur-rich protein,of which sulphur-containing amino acid amounts to 15.63% of the total amino acids.The chimeric plas- mid of 15kD zein gene regulated by rbcS promoter was introduced into Astragalus cicer by Agrobacterium mediated,and transformed plants have been obtained.NPTⅡ enzyme activity had been demonstrated in the kanamycin-resistant plantlets.The data from Southern blot indi- cated that the 15kD zein gene had been integrated into the chromosomal genome of Astragalus cicer.
