水稻rbcL基因的克隆及其结构分析

摘要 应用 M13mp18/19-JM101以及 pUC118/119-MV1184系统对籼稻(Oryza sativa L.,ssp.indica)rbcL 基因进行了分子克隆,并构建了一组有序缺失型。采用双脱氧末端终止法测定了籼稻叶绿体 rbcL 基因全顺序,结构基因全长1434bp,编码477个氨基酸。籼稻品种和粳稻品种仅在1056位有一个核苷酸的差异,但并不影响所编码氨基酸的种类。该基因5端启动子区和 atpB 基因5′端启动子十分接近,仅相隔159个核苷酸。而3′端下游区存在着与部分 rp123基因同源的重复性片段。我们还应用计算机软件对迄今为止已测的各种生物的 rbcL 基因进行了系统的分析比较。 The gene for the large subunit of ribulose 1,5-bisphospate carboxylase in rice(Oryza sa- tiva L.,ssp.indica)has been cloned using M13mp18/19-JM101 and pUC118/119-MV1184 sys- tems.After subcloning and construction of a series of unidirectional deletions,the nucleotide se- quence was determined by the dideoxychain termination method.The structure gene encom- passes 1434,which encodes 477 amino acids.The two subspecies of rice,i.e.japonica and indica have only one diverse nucleotide,which is located in the third site of a codon and does not change the kind of amino acids.The promoter region of rbcL gene is closely contiguous to that of atpB gene in its 5'-terminal upstream of rbcL gene.A repetitive sequence homologous to a part of rp123 gene exists in the 3'-terminal downstream of rbcL gene.With the aid of com- puter,we have compared the sequence of rbcL gene in rice with those in other species deter- mined so far to investigate the phylogenetic topologies.
