摘要
在植物电镜样品的超薄切片中,材料的定位很重要,特别是对那些用肉眼无法辨别的微小材料如胚囊及其内部的卵器、组织培养中的胚性细胞等则更困难。Kennell(1985)曾介绍一种重包埋技术,但该技术步骤多,需要时间长,操作较复杂。我们参考了Kennell的工作,对重包埋技术作了较大改进,解决了微小材料定位难的问题。
The Epon--812 semithin-section, which had been observed under light microscope, was stuck to a trimmed resin block wth 502 instant glue and was ultrasectioned for electron micro- scopy. This method, which is simple and repid, gives a good result and may solve some pro- blems happened in ultrasectioning some special material which needs to be exactly located.
