大鼠急性脊髓损伤后神经细胞凋亡的实验研究

摘要 目的 :研究细胞凋亡在急性脊髓损伤中的作用。方法 :以大鼠脊髓压迫性损伤的动物模型 ,应用透射电镜 ,HE染色 ,原位缺口末端标记 (TUNEL)观察急性脊髓损伤后神经细胞的死亡形式。结果 :急性脊髓损伤后 2h电镜清晰观察到神经细胞凋亡的形态变化 ,HE染色和TUNEL显示急性脊髓损伤后 6h和 8h凋亡阳性细胞达高峰。结论 :细胞凋亡为急性脊髓损伤后神经细胞的主要死亡形式 ,也是加重脊髓损伤的一个因素。 Objective: To investigate the mechanism of apoptosis in nerve cells after acute spinal cord injury (SCI). Methods:The transmission electron microscope(TEM), HE staining, terminal deoxynucleotidy1 transferase mediated dUTP nick end labeling(TUNEL) were used to evaluate the way by which the nerve cells died in acute spinal cord injury in the rat model. Results:Two hours after acute SCI, the morphological changes were clearly demonstrated by the TEM. Observed with the HE staining and TUNEL, the positive apoptotic cells reached the peak 6 to 8 hours after acute SCI. Conclusion:Apoptosis was the main pattern by which the nerve cells died in acute SCI, and it also was a factor in aggravating the injury of spinal cord.
