摘要
目的 :运用基因芯片技术研究曲张大隐静脉基因表达谱的变化。方法 :选取原发性大隐静脉曲张病人隐 股交界瓣膜区静脉 6条 ,取 6条正常静脉作对照。提取血管组织总RNA ,纯化mRNA后逆转录制备目标序列 ,应用含有 4 0 96种人类基因cDNA的表达谱芯片进行差异表达谱分析。结果 :在原发性大隐静脉曲张的基因表达谱中差异表达基因共有 16 8条 ,上调基因 96条 ,下调基因 72条 ,共存性差异表达基因 39条 ,其中上调基因 2 8条 ,下调基因 11条。差异表达基因中有细胞凋亡相关基因、原癌基因和抑癌基因、细胞信号和传递蛋白基因等多种基因。这些基因可能与原发性大隐静脉曲张的发病机制有关。结论 :采用基因芯片技术筛选大隐静脉曲张的致病相关基因 ,可能为其病因和发病机制的研究提供新思路。
Objective: To screen the differently expressed genes between varicose and non-varicose veins of great saphenous vein using cDNA microarray. Methods: Six veins with varicose and 6 normal veins near the saphenofemoral junction were collected, and total RNA were isolated and mRNA were purified by using oligotex. Both mRNAs from varicose veins and normal veins were reversely transcripted to cDNAs with fluorescent dUTP to prepare the target sequences, then the target sequences were hybridized to cDNA microarray of 4096 human genes. After washing, the cDNA microarray were scanned for the fluorescent signals and showed differences between 2 kinds of tissues. Results: Among 4096 gene probes, 168 genes showed differential expressed profile with 96 upregulated and 72 downregulated genes, and all 6 varicose samples had 28 upregulated and 11 downregulated genes. The differential expressed genes were related with apoptosis, proto- or anti-oncogene, cell signal conduct, and cytoskeleton, etc. These genes might be related with the pathogenesis of varicose. Conclusion: The investigations based on cDNA microarray can sequence related genes in varicose of great saphenous vein, and may provide a new idea in studying pathogenesis of varicose.
