颊拭子样本检测牙周炎相关基因的应用 被引量:5

摘要 目的 探讨采用颊黏膜拭子抽提DNA分析Fcγ受体 (FcγR)IIIA、基质金属蛋白酶 (matrixmetalloproteinase ,MMP) - 9基因多态性的可行性。方法 分别提取 10例慢性牙周炎患者静脉血和颊黏膜拭子DNA ,采用限制性内切酶酶切片断长度多态性 (PCR -RFLP)技术 ,检测FcγRIIIA - 15 8和MMP - 915 6 2基因多态性并进行比较。结果 颊黏膜拭子抽提DNA与静脉血抽提DNA用于分析FcγRIIIA、MMP - 9基因多态性的结果完全一致。结论 颊黏膜拭子具有无创、方便的特点 ,所抽提DNA适于FcγRIIIA、MMP - Objective To develop a rapid genotyping method detecting FcγRⅢA and matrix metalloproteinase-9 (MMP-9) from buccal swabs.Methods DNA was extracted from blood leucocytes and buccal swab samples from 10 subjects with chronic periodontitis.Genotypes of the FcγRⅢA and MMP-9 were determined with PCR-RFLP respectively.The difference of each genotype between those two methods was compared.Results No differences in the quality of amplification or fragment separation in the electrophoresis were observed.Conclusion FcγRⅢA and MMP-9 genotypes can be determined from DNA extracted from buccal swabs.The buccal swab is a simple, non-invasive and high patient acceptance method to get DNA for studies of Fcγ receptor ⅢA and MMP-9 polymorphisms.
