力竭运动后恢复不同时程骨骼肌中MMP-2的表达 被引量:1

Study on the Expression of MMP-2in Mice' s Akeletal Muscle at the Different Recovery Time Course After the Exhausted Swimming

摘要 目的:探讨游泳力竭运动后恢复不同时程基质金属蛋白酶-2(MMP-2)在小鼠小腿三头肌中的分布和表达。方法:制备小鼠游泳力竭运动模型,用免疫组织化学法研究小鼠游泳力竭运动后恢复不同时程小鼠小腿三头肌中MMP-2的分布和表达,并用计算机图像分析系统对结果进行分析。结果:(1)力竭运动后,恢复不同时程MMP-2在小鼠小腿三头肌中有不同程度的表达,阳性部位主要分布在慢肌肌纤维和肌内膜,快肌纤维肌膜内侧有较弱的免疫阳性反应;(2)游泳3h力竭运动后即刻,小腿三头肌中MMP-2表达量明显高于对照组(P<0.01);(3)游泳3h力竭运动后恢复1、6、12h,小腿三头肌中MMP-2表达量仍明显高于对照组(P<0.05)。其中在恢复6h时出现了MMP-2的表达高峰,与对照组相比差异显著(P<0.01);(4)随着恢复时间的延长,MMP-2表达量逐渐下降,恢复24h时MMP-2表达量已与对照组间无显著性差异。结论:力竭运动导致MMP-2的表达增加,而且在恢复过程中,骨骼肌中MMP-2表达具有时效性,这种表达可能与力竭运动后产生的骨骼肌过度疲劳和损伤有关。 Purpose:To approach the distribution and expression of MMP-2in the triceps of the mice' s shank at different recovery time course after3h exhausted swimming exercise.Methods:42male Kunming mice were subjected to forced swimming exhausted exercise,the distri-bution and expression of MMP-2mentioned above were examined by the method of immunohistochemistry,then were analyzed the results were analyzed through the analysis system of computer picture.Results:(1)After exhausted exercises,MMP-2had different expression pattern at the different recovery time course.Positive expression was mainly distributed at the muscle fiber of slow muscle and endomysi-um,and there was a weaker positive expression in the interior of the membrane of fast muscle fiber;(2)At the moment of3h exhausted ex-ercise later,the expression quality of MMP-2in the triceps of the mice' s shank was obviously higher than that of the control group(P<0.01);(3)At the first,the sixth and the twelfth hour of the recovery time course after the exhausted exercises,the expression quality of MMP-2was still obviously higher than that of control group(P<0.05).Especially,a expression peak of MMP-2appeared at the sixth hour of the recovery time course,and had a significant difference compared with the control group(P<0.01);(4)With the prolongation of recov-ery time course,the expression of MMP-2descented gradually.At the twenty-forth hour,there was no obvious difference between the ex-perimental group and the control group.Conclusion:Exhausted exercise adds up to the expression quality of MMP-2,and which in skeletal muscle has time-effect in the course of recovery.This expression pattern may be related to the overfatigue and injury of skeletal muscle af-ter exhausted exercise.
