CD44在大鼠肾小球硬化模型中的表达

摘要 目的 探讨CD44在大鼠进行性肾小球硬化中的作用。方法应用免疫组化、原位杂交及彩色病理图像分析系统观察柔红霉素所致的大鼠进行性肾小球硬化模型不同时相肾组织中CD44及其mRNA的表达,分析其与进行性肾小球硬化病理形态学改变的关系。结果 大鼠肾小球硬化过程中肾小球内CD44及其mRNA表达增高,主要位于内皮细胞、足细胞、肾小囊上皮细胞以及系膜细胞。肾小球内细胞平均CD44阳性面积和肾小球内CD44 mRNA阳性细胞百分率显著增高,且均与肾小球硬化指数正相关(r1=0.754,r2=0.744)。结论 CD44高表达可能在促进肾小球硬化中发挥重要的作用。 Objective To clarify the possible role of CD44 in the pathogenesis of progressive glomerulosclerosis in rats. Methods The glomerulosclerosis model in rat was established by injecting daunomycin. Immunohistochemistry.in situ hybridization and colour pathological image analysis system were used to evaluate the relationship between CD44 expression and the progressive pathological changes of glomerulosclerosis at different time. Results CD44 and its mRNA expressions of glomerulosclerosis increased markedly in endothelial cells,podocytes,epithelial cells of Bowman's capsule,me-sangial cells. CD44 positive area per cell and the percentage of CD44 mRNA positive cells in glomeruli increased in progressive glomerulosclerosis. They correlated positively with glomerular sclerosis index (r1 =0. 754,r2 =0. 744). Conclusion CD44 may play a pivotal role in pathogenesis of progressive glomerulosclerosis.
