摘要
目的:检测肿瘤坏死因子相关凋亡诱导配体(TRAIL)的受体在正常人外周血单个核细胞上的表达,并探讨其意义。方法:采用半定量逆转录聚合酶链反应(RT-PCR)和流式细胞术,对TRAIL受体的表达分别进行RNA水平和蛋白水平的检测。结果:各受体经RT-PCR和流式细胞术检出的阳性率分别为DR4(50%、60%)、DR5(100%、100%)、DcR1(100%、100%)、DcR2(60%、70%),其中死亡受体DR5和诱骗受体DcR1的表达阳性率最高,均为100%。RT-PCR和流式细胞术均显示:能够诱导凋亡反应的死亡受体中,DR5表达显著高于死亡受体DR4P<0.05;能够竞争性阻断TRAIL诱导的凋亡反应的诱骗受体中,DcR1表达显著高于诱骗受体DcR2P<0.05。结论:DR5和DcR1是外周血单个核细胞PBMC上TRAIL受体系统的主要成员,在TRAIL诱导的细胞凋亡中起关键作用。
Objective: To detect the expression of TRAIL receptors on PBMC of healthy people and explore their significance. Methods: The expressions of TRAIL receptors were assayed by semiquantitive RT-PCR and flowcytometry. Results: Among all of the four receptors, the expression of DR5 is much higher than that of DR4(P<0.05); DcR1 is far higher than DcR2 (P<0.05). DR5 and DcR1 have the highest positive rate 100%. Conclusions: DR5 and DcR1 may play a key role in the apoptotic reaction of PBMC induced by TRAIL.
