二价金属阳离子对牙龈卟啉单胞菌FtsZ的GTPase活性的调控及意义 被引量:2

摘要 目的 :探讨二价金属阳离子对牙龈卟啉单胞菌FtsZ的GTPase活性的影响。方法 :将质粒 pEZ1(WtPgFt sZ ,野生型PgFtsZ)、pYW 1(ZΔC0 1,PgFtsZ的C末端缺失 73个氨基酸残基的变异型 )和pYW 2 (ZΔC0 2 ,PgFtsZ的C末端缺失 12 8个氨基酸残基的变异型 ,已去除了PgFtsZ的C末端非保守性的区域 )导入EscherichiacoliBL2 1(DE3)pLysS中表达 ,然后进行分离和纯化这些目的蛋白。使用Malachitegreenassay法 ,检测 10mM /L的各种二价金属阳离子对纯化的WtPgFtsZ、ZΔC0 1和ZΔC0 2的GTPase活性的作用。 结果 :二价金属阳离子对WtPgFtsZ、ZΔC0 1和ZΔC0 2的GTPase的活性作用的特性相似 ,既在 10mM /L的Mg2 + 存在下与没有Mg2 + 的情况下它们的GTPase的活性相似 ;然而 ,在 10mM /L的Ca2 + 、Mn2 + 、CO2 + 和Ni2 + 存在的情况下WtPgFtsZ、ZΔC0 1和ZΔC0 2的GTPase的活性表现出不同程度的减弱 (36 .7%~ 70 .8% )。结论 :10mM /L的Mg2 + 对WtPgFtsZ、ZΔC0 1和ZΔC0 2的GTPase的活性无影响 ,而 10mM /L的Ca2 + 、Mn2 + 、CO2 + 和Ni2 + Objective:To investigate effect of divalent metals on the GTPase activity of FtsZ in Porphyromonas gingivalis.Methods:Plasmids pEZ1 (Wt PgFtsZ), pYW1(ZΔC01, missing 73 residues from the C-terminus of PgFtsZ) and pYW2 (ZΔC02, missing 128 residues from the C-terminus of PgFtsZ) were introduced into Escherichia coli BL21(DE3)pLysS to express and purify the purpose proteins.Malachite green assay was used to measure the GTPase activities of purified Wt PgFtsZ, ZΔC01 and ZΔC02.Results:GTPase activities of Wt PgFtsZ, ZΔC01 and ZΔC02 were not affected by 10 mM Mg 2+.However, 10 mM others divalent metals including Ca 2+, Mn 2+, Co 2+and Ni 2+ reduced the GTPase activities of Wt PgFtsZ, ZΔC01 and ZΔC02(36.7% ~70.8%).Conclusions:Effects of divalent metals on GTPase activities of Wt PgFtsZ, ZΔC01 and ZΔC02 have similar characterizations, viz,10 mM Mg 2+ has no effect on GTPase activities of Wt PgFtsZ, ZΔC01 and ZΔC02. 10 mM Ca 2+, 10 mM Mn 2+, 10 mM Co 2+ and 10 mM Ni 2+ inhibit the GTPase activities. Furthermore, the non-conserved C-terminal region of PgFtsZ is not need for the effect of divalent metals on the GTPase activity.
