饥饿的盘基网柄菌进入多细胞发育期 ,在发育早期 ,AK12 7细胞 (gp150突变细胞 )能表达DdCAD 1和 gp80两种粘附分子 ,但它们不足以促进细胞继续发育 ,发育停留在细胞疏松结合阶段。粘附分子 gp150调节的细胞与细胞间的粘着影响了细胞丘“突出”的形成 ,由此影响了盘基网柄菌多细胞发育的形态发生。TL93细胞 (DdCAD 1和gp80突变细胞 )能完成发育。主要原因是在细胞流发育阶段就表达了gp150分子 ,在细胞粘着的功能上有替代DdCAD 1和 gp80的作用。因此
Cells of Dictyostelium discoideum are triggered to undergo development when they have depleted their food source. During the early hours of developmen t, cells undergo chemotaxis toward cAMP that is secreted by starving cells and f orm multicellular aggregates, which finally develop into the fruiting bodies. Ce ll adhesion molecules play an important role in maintaining the on integrity of cell aggregation and influence subsequent cell-type differentiation and cell so r ting. Cell-cell adhesion at early stages is important for development, but cel l -type differentiation and cell sorting are triggered in the mound stage. Mutant s lacking the gp80 gene exhibit no obvious difference in phenotype when they deve lop under standard conditions. Perhaps related proteins make compensation for t he mutant. gp150 is normally first expressed at the mound stage so the question is whether the adhesion molecule is important for development beyond this stage ? To better understand the role of gp150, we compared mutant cells and wild-t y pe cell with Western blot and immunofluorescence labeling. AK127 cell (gp150/Lag C null cell) expressed both DdCAD-1 and gp80 on the cell surface during early d e velopment, arrested at the loose aggregation stage. gp150 that regulated the cel l aggregation, influenced the formation of the tip of mound. TL93 cells have no obvious difference in phenotype when developed under standard laboratory condit ion. Premature change in gp150 gene expression was observed, which perhaps compe nsate the other related adhesion proteins. gp 150 is required for the developmen t of Dictyostelium discoideun .
