平肝熄风汤对脑出血大鼠海马线粒体膜电位和神经功能的影响(英文)

Effect of Pinggan Xifeng Tang on membrane potential of hippocampal mitochondria and neurological function following intracerebral hemorrhage in rats

摘要 目的:观察大鼠脑出血后神经缺损症状和线粒体膜电位的改变,及中药制剂平肝熄风汤的干预作用。方法:用Ⅶ型胶原酶于脑内定位注射诱导大鼠脑出血模型。神经缺损积分评定神经功能;用荧光分光光度计测定线粒体悬液的荧光强度反映线粒体膜电位。结果:大鼠脑出血后迅速出现神经缺损症状,平肝熄风汤能明显降低神经缺损症状积分(4h:8.80分,1d:6.80分,3d:2.80分,7d:1.60分)。脑出血大鼠线粒体膜电位于术后4h显著下降,1d时膜电位下降达高峰,3,7d逐步恢复,仍低于正常组和假手术组(t=7.352P<0.05)。平肝熄风汤治疗组在脑出血后1,3,7d,线粒体膜电位均明显高于模型组(t=2.583P<0.05)。结论:大鼠脑出血后线粒体膜电位下降;平肝熄风汤能阻断膜电位下降,同时能减轻出血后神经损伤。 AIM:To observe the symptoms of neurological impairment induced by cerebral hemorrhage and the changes of mitochondrial membrane potential so as to investigate the curative effect of Pinggan Xifeng Tang.METHODS:Cerebral hemorrhage models of rats were established by stereotaxical injection of collagenaseVII into the pallidum.Neurological function was scored by neurological impairment scale.Fluorescent intensity of mitochondrial suspension was determined with spectrofluorometer for testing the membrane potential.RESULTS:When the rats had cerebral ischemia, there quickly appeared the symptom of nerve damage.Pinggan Xifeng Tang functioned clearly to decrease the scores of neurological impairment(4 h:8.80 scores,1 d:6.80 scores,3 d:2.80 scores,7 d:1.60 scores).The membrane potential of mitochondria decreased greatly 4 hours after operation,decreased to the lowest level one day after operation,and restored gradually at 3-7 days after operation, but was still lower than that of the normal and pseudo-operation groups (t=7.352, P< 0.05).The mitochondrial membrane potential of the Pinggan Xifeng Tang group was obviously higher than that of the model group 1,3,and 7 days after ischemia , respectively(t=2.583,P< 0.05). CONCLUSION:The mitochondrial membrane potential of the cerebral ischemiac rats decreases;Pinggan Xifeng Tang can inhibit the decrease, and at the same time, relieve the nerve damage.
