摘要
介绍观察被子植物胚囊结构的方法。现较常用的方法有整体染色切片法在普通光学显微镜下观察;整体染色透明法或整体透明法在相差显微镜、微分干涉差显微镜或激光共聚焦扫描显微镜下观察;酶解法分离胚囊在微分干涉差显微镜、荧光显微镜或普通光学显微镜下观察。文章并讨论了各种观察方法的优缺点。
This paper introduced some easy and rapid procedures for observation of embryo sac structure of angiosperm, including whole stain-section technique with the use of optical microscope,whole stain-clearing technique or whole clearing technique with the use of phase contrast microscope,differential interference contrast microscope or confocal laser scanning microscope and embryo sac isolation by means of enzymatic maceration technique with the use of differential interference contrast microscope, fluorescence microscope or optical microscope.And,their advantages and disadvantages were emphasized.
