氯气染毒大鼠血浆中肿瘤坏死因子的变化及丹参的影响

摘要 目的 检测氯气染毒致肺水肿时大鼠血浆中肿瘤坏死因子 (TNF α)的变化及丹参对这种变化的影响 ,探讨此种变化与肺水肿的关系以及丹参的可能治疗机制。方法 利用氯气染毒大鼠致肺水肿 ,治疗组大鼠经静脉注入丹参注射液 ,以放免技术测定各组血浆中TNF α含量 ,测定各组肺干重和湿重并计算肺含水量。结果 染毒组大鼠血浆中TNF α含量及肺含水量均显著高于对照组 (P <0 0 0 1) ;丹参治疗组大鼠血浆中TNF α含量及肺含水量均显著低于染毒组 (P <0 0 0 1)。结论 氯气染毒可导致肺水肿大鼠TNF α含量显著升高 ,这可能是发生肺水肿的机制之一。丹参可降低血中TNF α浓度 。 Objective This study was designed to investigate changes in plasma concentration of tumor necrosis factor-α(TNF-α)in rats with pulmonary edema caused by chlorine gas exposure and effect of radix salviae miltiorrhizae on it,and to explore possible mechanism of radix salviae miltiorrhizae treatment.Method Rat model with pulmonary edema was prepared by exposure to chlorine for 10 minutes.The rats were randomized into groups of control and treatment with intravenous injection of radix salviae miltiorrhizae.Plasma level of TNF-α in the rats was measured with radioimmunoassay,their wet and dry weight of the lung was determined and its water content was calculated.Result Plasma level of TNF-α and water content of the lung in rats exposed to chlorine were significantly higher than those of controls(P<0.001).Plasma level of TNF-α and water content of the lung in the exposed rats treated with radix salviae miltiorrhizae were significantly lower than those without treatment(P<0.001).Conclusion Chlorine gas could cause plasma level of TNF-α increased in exposed rats with pulmonary edema,which was possibly one of its mechanisms.Radix salviae miltiorrhizae could lower plasma level of TNF-α in rats and reduce their pulmonary edema.
