摘要
目的 :研究MCI 186对PC12细胞凋亡的影响。方法 :以过氧化氢诱导的PC12细胞凋亡模型 ,采用MTT比色法 ,琼脂糖凝胶电泳分析 ,荧光染色及荧光显微镜形态学观察 ,流式细胞术 (FCM)检测细胞凋亡。结果 :MCI 186可有效地改善凋亡引起的PC12细胞形态学变化 ,提高细胞存活率 ,1× 10 -5mol/LMCI 186可减小亚二倍体峰的峰面积 ,使凋亡细胞比率 (1 99)小于损伤对照组细胞 (6 45)。结论 :MCI 186可能作用于细胞凋亡过程 ,通过清除活性氧 。
AIM: To examine the effect of MCI-1 86 on neuronal apoptosis METHOD: Using PC12 cells as the mode l of neur ons,in addition to MTT method,fluorescence microscopy,agarose gel electrop hor esis and flow cytometry,the apoptosis induced by H 2O 2 and the effec t of MCI-186 on PC12 cells were observed RESULT: In PC12 cell culture,MC I- 186 at 1×10 -5 mol/L was able to significantly attenuate changes of cell morphology ind uced by H 2O 2,and improve the survival Moreover,the ratio of ap optosis was decreased from 645 in control group to 199 CONCLUSION: Th e data indicate that MCI-186 inhibits apoptosis induced by H 2O 2 and the neuroprotective effects of MCI-186 are associated with its anti-apoptosis effe ct s by reducing ROS production However,the precise role of MCI-186 in apoptot ic pathway and its intracellular signaling cascade remains to be examined furthe r
