抗氧化微量营养素对糖尿病小鼠细胞因子基因表达的调控作用 被引量:4

摘要 目的 研究抗氧化微量营养素 (Antioxidantmicronutrients,AM)对糖尿病小鼠细胞因子基因表达调控作用 ,寻找AM作用的敏感基因 ,探索防治糖尿病的分子生物学策略。方法 通过腹腔注射 2 %四氧嘧啶制备IDDM模型 ,经灌胃方法联合给予硒 (Se)、维生素E(VE)、钒 (V)和铬 (Cr) ,应用流式细胞仪测定外周血淋巴细胞表达IL 2、IL 10、IFN γ和TNF α细胞数百分比以及脾活性氧 (ROS)含量。结果 IDDM模型组小鼠外周血表达IL 2和IL 10水平明显低于AM组和正常组 (P <0 0 1) ;而表达IFN γ水平明显高于其他两组 (P <0 0 1) ;各组表达TNF α无明显差异 (P >0 0 5 ) ;IDDM模型组脾ROS含量明显增高 (P <0 0 1) ,AM组脾ROS含量较模型组明显降低 (P <0 0 1)。结论 联合应用AM可上调糖尿病小鼠IL 2、IL 10基因表达 ,下调IFN γ基因表达 ,并可降低糖尿病小鼠脾中ROS含量 ,从而起到拮抗糖尿病的作用。 Objective To investigate the regulatory mechanism of antioxidant micronutrients( AM) on the expression of cytokines genes in diabetic mice and find the sensitive genes which AM affects, and explore the molecular biological strategies of preventing from and treating for diabetes. Methods IDDM models were made by the intraperitoneal injection of 2% Alloxan. Taking Selenium(Se), Vitamine E(VE), Vanadium(V) and Chrome(Cr) orally everyday, then the percentage of IL-2,IL-10, IFN-γ and TNF-α positive blood lymphocytes as well as ROS in spleen of mice were detected by flow cytometer (FCM). Results Compared with AM group and control group, the percentage of IL-2 and IL-10 positive blood lymphocytes of model group was significantly decreased(P<0.01),while the percentage of IFN-γ positive blood lymphocytes and ROS contents in spleen of model group were increased(P<0.01);There was no significant difference in expressing TNF-α among these groups.Conclusion Combined supplement of AM could upregulate the expression of IL-2 and IL-10 genes of diabetic mice and decrease IFN-γ gene expression and decrease contents ROS in spleen ,then diabetes of mice could be effectively treated.
